Yes. Gyms are one of the side activities Rockstar confirmed to Game Informer, and the Extended Look shows Lucia working out in a button-press minigame. Regular workouts make Jason and Lucia visibly stronger and more muscular.
Gyms sit on Rockstar's activity list next to base jumping, billiards, kayaking, mini golf and scuba diving, so they are a regular part of free roam rather than a one-off mission.
The gym ties into two other systems. Eating adds weight and restores health, while working out builds muscle and strength. Training together with your partner also counts toward Jason and Lucia's relationship.
IGN compares the gym minigame to GTA San Andreas. Rockstar has not said exactly what extra strength does in GTA 6, beyond the change in appearance and the strength boost IGN describes.
